Hi mate,
I had the exact same issue this week. Hooked the pump up to a temp hose and worked perfect (phew). Put a new plug in the vertical cylinder and bike fired up first crank.
As many have already said hopefully it’s just the vertical as the horizontal is more involved.

NGK spark plugs ?, they behave like you description.
Like others write replace them.
You can save the old plugs with a gentle glassblowing , but replacement is better.
Did you replace with same plugs, or do you recommend a different brand?
 
I believe if you try to put anything else in it it’s either gonna be too short or the piston will make contact with it

Dude had great idea - to try swapping out just the vertical and see if it will start. Front plug is huge pita to get to

Take it the ether didn’t work?
 
I love that the manual says just remove the little access plate to change the front plug, but you must have to have italian fingers to actually get the cap off! Ether down the inlets was not sufficient. Thought if I was accessing direct, may as well do at least rear plug.

For the Pani I mean you only can get some from NGK
Had the same problems with my 998 and my HM1100, changede to Champion and newer had problems after that.
Worst experience I ever had with NGK, was with a Suzuki GSX-R250 (4 cyl. with 45HP, needed to be rum over 8000 rpm all time, but my wife often ran it below ), the spark plougs only lasted for a few weeks then they needed a glassblowing before the could again, changde to a another brand and newer had problems again.
 
Hey Paul don’t be bummed man. It ran after you got home and it will run again soon. I noticed over the years more than a few of the race bikes I saw were set up with a ‘cold start plug’. The guys would hook them to a jump box (not to a trickle charger) for the initial fire each day. Granted they all had light weight lith ion batt but I am thinking you are not the first guy to get bit by this. Yeah I still think it’s just plugs. Let’s hope I am right. I know this is a lil sucky for you at the moment but it has inspired me to add on more thing to my list that might keep me from having problems later. Thanks : )

Thanks. I'm hoping to get the plugs and a spark plug tool today (of course none of the spark plug tools I have will fit!) so I can have a crack and hopefully get it fired up. I'm sure once I have it running smoothly I'll love it. I actually have a portable jumper box, so I may even go overboard and hook that up from the outset to give it the best chance of success.
 
There is no injector relay under the seat. The coils are pretty stout they rarely go bad also the spark cables as well. Easy check is just pull the vertical spark plug make sure its grounded to the case turn motor over should see spark. Best thing is pull the tank make sure you got fuel in the line to the injectors. I made a extension harness and extension fuel line so I can run the bike with tank off the bike makes it easy to diagnose stuff. Injectors don't open up till the bike fires so if you see wet fuel in the throttle bodies and on spark plug your getting fuel. If a MAP sensor or anything on the 5v circuit is acting up bike wont run.
 
Also was the bike back firing at all out the exhaust? If its backfiring out the intake would be bigger problems so if you are getting fuel, spark and its backfiring I would be swapping MAP sensors. Even if your getting spark on one cylinder it will still run so anyway you got few things go check.
 
Thanks team. New plug fitted in rear/vertical cylinder, battery charged and I allowed the garage to be warmed up (wife was not so happy about having the door open all day :) ) and bike started first attempt. Took it out for a good spin, no problems at all. Loved it!
